Project Overview
CD3 bispecific T cell engagers (TCEs) are antibody-based therapeutics that binding both to a tumor associated antigen and CD3 on T cells, thereby directing the T cells to the tumor cells and activating them to kill the tumor. In preclinical development, mechanistic PK/PD models or QSP models are commonly used to predict the dose-exposure-response relationship and to project the pharmacologically active dose range in patients. Commonly, such PK/PD models for TCEs characterize the formation of ternary complexes (trimers) between T cell, antibody and target cell to link antibody concentrations to pharmacological activity (T cell activation, expansion, cytokine release, cytotoxicity). Models of varying complexity have been described in literature. However, the experimental work required to calibrate these models may be substantial, e.g. in vitro T cell mediated cytotoxicity experiments across multiple tumor cell lines, T cell donors, effector-to-target ratios, antibody concentrations and time points.
As an intern in the Quantitative Translational Pharmacology team at Genmab, you will contribute to developing a generic translational strategy for TCEs to make the early stages of drug development more efficient. Your responsibilities will include an evaluation of various TCE PK/PD models, their performance in describing the preclinical data and predicting clinical dose-response. Your recommendations for a translational modeling strategy that maximizes predictive value while minimizing experimental work required for model calibration will be of great value in speeding up the timelines of bringing candidate TCEs from discovery to patients.
You will work with diverse types of data, including molecular and cellular binding, T cell mediated cytotoxicity In vitro, nonclinical PK, tumor growth inhibition in xenograft mouse models, clinical PK and dose-response data.
You will gain practical experience in data handling, quantitative modeling and scientific communication within a dynamic biotech environment and experience the impact of modeling and simulation on key drug development decisions.
